Optimization of ultrasonication-assisted extraction conditions using RSM-I-Optimal experimental design to recover vitamin D2 and K1 from selected green leafy vegetable samples

This study employed the response surface methodology to optimize extraction conditions for recovering vitamins D2 and K1 from green leafy vegetables using ultrasonication-assisted extraction. The vitamin content was determined an Accucore C18 column a UPLC-Q-ToF/MS method. An RSM-I-Optimal design used designing experiment find best combination of solvent level (mL), sonication time (min), frequency (kHz), temperature (°C). experimental data 25-sample set were fitted second-order polynomial equation multiple regression analysis. models had R2 values 0.895 0.896, respectively, probability (p < 0.0001) indicated that model highly significant. optimal were: 65 mL, 45 min, 70 kHz, °C. Under these conditions, predicted recovery (%) 90.7% 90.4%, respectively. has potential use reported method routine quantification in laboratory UPLC-Q-ToF/MS.
